Summary A theoretical calculation is presented which extends Wright's island model of drift and migration to differential migration between the two sexes. In this circumstance, local demes no longer have Hardy-Weinberg frequencies. There may be local heterozygote excess or deficiency depending, respectively, on whether migration occurs before or after mating. The magnitude of the local departure from Hardy-Weinberg is directly proportional to the difference between the migration parameters of the two sexes. These results could have important implications for studies where genetic markers are used for inferring population structure. An example from a study of Marmot colonies is cited. 相似文献

Levels were evaluated of Aflatoxin B1 (AFB1) exposure through the intake of AFB1-contaminated foods among Koreans. Average daily intake, excess cancer risk, and population risk of AFB1 from food intake were calculated from Koreans with age greater than 20 years. Average daily intake of AFB1 by food ingestion was 0.064 ~ 0.361 ng/kg bw/ day. Excess cancer risk of AFB1 in food with the surface antigen of the hepatitis B-virus negative (HBsAg?) and positive (HBsAg+) population was 5.76 × 10? 7~ 3.25 × 10? 6 and 1.47 × 10? 5~ 8.31 × 10? 5, respectively. Application of JECFA's two tentative maximum levels (MLs) (10 μ g/kg and 20 μ g/kg) resulted in the population risk of 0.0009 ~ 0.0075 and 0.0014 ~ 0.0079 cancers/year/100,000 people, respectively, indicating that lower ML does not give a significant reduction of the risk. However, because Korea has a high incidence of HBsAg+ individuals, adoption of lower ML may be a reasonable decision to protect a high risk group. This is the first report on the risk assessment of AFB1 in food among the Korean population. Our study will contribute to providing the solid basis of establishing the maximum levels of AFB1 exposure through food ingestion in Korea as well as other countries. 相似文献

ObjectiveIodine is essential for thyroid hormone production. The recommended dietary allowance for iodine in nonpregnant adults is 150 μg/d. However, most product labels do not list the iodine content. Meal replacements are not required to contain specific vitamins and minerals. Nevertheless, they are often marketed as good and convenient sources of a balanced nutrition. In this study, we aimed to assess the iodine content in meal replacements to determine how they may contribute to iodine deficiency or excess.MethodsTwenty seven meal replacements from supermarkets in the Boston area were collected. The iodine concentration of each meal replacement was measured spectrophotometrically. Iodine content in meal replacements were compared according to form (liquid, bar, and powder) and type (vegan and nonvegan).ResultsThe overall mean ± SD iodine content was 49.7 ± 125.4 μg/serving. However, 1 meal replacement was an outlier and had 671.9 μg iodine per serving. Mean iodine content differed between forms and was highest for liquids (mean ± SD: 37.4 ± 6.5 μg/serving). Nonvegan meal replacements had a higher mean iodine content than vegan meal replacements (mean ± SD: 31.6 ± 15.78 μg/serving).ConclusionAll of the meal replacements contained detectible amounts of iodine regardless of whether it was listed on their labels (41% did not list iodine). Overall, the meal replacements in this study were found to be good sources of iodine. Coordinated lipid metabolism contributes to maintaining skin homeostasis by regulating skin barrier formation, immune reactions, thermogenesis, and perception. Several reports have documented the changes in lipid composition in dermatitis, including in atopic dermatitis (AD); however, the specific mechanism by which these lipid profiles are altered during AD pathogenesis remains unknown. Here, we performed untargeted and targeted lipidomic analyses of an AD-like dermatitis model resulting from constitutive activation of Janus kinase 1 (Spade mice) to capture the comprehensive lipidome profile during dermatitis onset and progression. We successfully annotated over 700 skin lipids, including glycerophospholipids, ceramides, neutral lipids, and fatty acids, many of which were found to be present at significantly changed levels after dermatitis onset, as determined by the pruritus and erythema. Among them, we found the levels of ceramides composed of nonhydroxy fatty acid and dihydrosphingosine containing very long-chain (C22 or more) fatty acids were significantly downregulated before AD onset. Furthermore, in vitro enzyme assays using the skin of Spade mice demonstrated the enhancement of ceramide desaturation. Finally, we revealed topical application of ceramides composed of nonhydroxy fatty acid and dihydrosphingosine before AD onset effectively ameliorated the progression of AD symptoms in Spade mice. Our results suggest that the disruption in epidermal ceramide composition is caused by boosting ceramide desaturation in the initiation phase of AD, which regulates AD pathogenesis. 相似文献

Water-mediated hydrogen exchange (HX) processes involving the protein main chain are sensitive to structural dynamics and molecular interactions. Measuring deuterium uptake in amide bonds provides information on conformational states, structural transitions and binding events. Increasingly, deuterium levels are measured by mass spectrometry (MS) from proteolytically generated peptide fragments of large molecular systems. However, this bottom-up method has limited spectral capacity and requires a burdensome manual validation exercise, both of which restrict analysis of protein systems to generally less than 150 kDa. In this study, we present a bottom-up HX-MS2 method that improves peptide identification rates, localizes high-quality HX data and simplifies validation. The method combines a new peptide scoring algorithm (WUF, weighted unique fragment) with data-independent acquisition of peptide fragmentation data. Scoring incorporates the validation process and emphasizes identification accuracy. The HX-MS2 method is illustrated using data from a conformational analysis of microtubules treated with dimeric kinesin MCAK. When compared to a conventional Mascot-driven HX-MS method, HX-MS2 produces two-fold higher α/β-tubulin sequence depth at a peptide utilization rate of 74%. A Mascot approach delivers a utilization rate of 44%. The WUF score can be constrained by false utilization rate (FUR) calculations to return utilization values exceeding 90% without serious data loss, indicating that automated validation should be possible. The HX-MS2 data confirm that N-terminal MCAK domains anchor kinesin force generation in kinesin-mediated depolymerization, while the C-terminal tails regulate MCAK-tubulin interactions. 相似文献

Spike glycoprotein of SARS-CoV-2 mediates viral entry into host cells by facilitating virus attachment and membrane fusion. ACE2 is the main receptor of SARS-CoV-2 and its interaction with spike has shaped the virus’ emergence from an animal reservoir and subsequent evolution in the human host. Many structural studies on the spike:ACE2 interaction have provided insights into mechanisms driving viral evolution during the on-going pandemic. [Purpose]This pilot study aimed to develop a regression model to estimate the excess post-exercise oxygen consumption (EPOC) of Korean adults using various easy-to-measure dependent variables.[Methods]The EPOC and dependent variables for its estimation (e.g., sex, age, height, weight, body mass index, fat-free mass [FFM], fat mass, % body fat, and heart rate_sum [HR_sum]) were measured in 75 healthy adults ( 31 males, 44 females). Statistical analysis was performed to develop an EPOC estimation regression model using the stepwise regression method.[Results]We confirmed that FFM and HR_sum were important variables in the EPOC regression models of various exercise types. The explanatory power and standard errors of estimates (SEE) for EPOC of each exercise type were as follows: the continuous exercise (CEx) regression model was 86.3% (R2) and 85.9% (adjusted R2), and the mean SEE was 11.73 kcal, interval exercise (IEx) regression model was 83.1% (R2) and 82.6% (adjusted R2), while the mean SEE was 13.68 kcal, and the accumulation of short-duration exercise (AEx) regression models was 91.3% (R2) and 91.0% (adjusted R2), while the mean SEE was 27.71 kcal. There was no significant difference between the measured EPOC using a metabolic gas analyzer and the predicted EPOC for each exercise type.[Conclusion]This pilot study developed a regression model to estimate EPOC in healthy Korean adults. The regression model was as follows: CEx = -37.128 + 1.003 × (FFM) + 0.016 × (HR_sum), IEx = -49.265 + 1.442 × (FFM) + 0.013 × (HR_sum), and AEx = -100.942 + 2.209 × (FFM) + 0.020 × (HR_sum). 相似文献

Abstract. The authors examine the isotopic composition of leaf water, at natural abundance levels, as influenced by transpiration rate. The isotopic composition of water of wheat leaves ( Triticum aestivum L. var. Aroona) was followed while their transpiration rate adjusted to 'steady-state' environmental conditions. Leaf diffusive resistance was modified by short-term salt treatment and by plant culture in either nutrient solution, free-draining sand, or vermiculite. Resultant changes in 18 O and 2 H in leaf water are described and fitted to the model of Leaney et al. (1985). The treatments with lower transpiration rates were found to have a greater fraction of their leaf water equilibrated with water vapour in the atmosphere. Comparable results were obtained with both 18 O and 2 H, with some differences being interpreted in terms of turbulence in the vapour diffusion path. The fraction of the leaf water equilibrated with the atmosphere varied between leaves of different ages. However, this may have been due to their different positions in the canopy. 相似文献

Aqueous dispersions of dimyristoyl phosphatidylcholine (DMPC) mixed with 1-palmitoyl-2-oleoyl phosphatidylcholine (POPC) or with 1-palmitoyl-2-stearoyl phosphatidylcholine (PSPC) have been studied by differential scanning calorimetry. Analysis of phase diagrams of the resulting bilayers indicated that both POPC-DMPC were miscible, but not ideally mixed, in the gel and in the liquid-crystalline states. In both states, estimated values for excess interaction energies were more positive for PSPC-DMPC than for POPC-DMPC, indicating that mixing was less ideal in the former system. 相似文献
